Imaging Cores - Optical

 

Overview of Services

If you are interested in biological electron microscopy, we invite you see our colleagues at the Imaging Cores - Electron.

The Imaging Core - Optical Core Facility is available to help researchers with light microscopy-based research projects. This core supports a wide range of microscopy techniques from basic research needs to more advanced techniques. The core’s co-managers have over 8 decades of combined microscopy expertise and are available to assist you with the best solution to your research problems, as well as to train users on all of the microscopes.

The Marley building location includes:

  • A Zeiss LSM880 34 channel Laser Scanning Confocal Microscope. This system is based on a Zeiss Observer Z1 inverted microscope, which is a laser scanning confocal microscope with seven laser lines, motorized stage, and temperature control capability.
  • A second Zeiss LSM880 system with a 34 channel detector is based on a Zeiss AxioImager Z1 upright microscope, which includes six laser lines, motorized stage, Zeiss Fast Airyscan super-resolution attachment and temperature control as well as a Spectra-Physics MaiTai femtosecond pulsed laser for multi-photon imaging.
  • A Zeiss Axiozoom fluorescent stereo microscope equipped with a Hamamatsu Flash 4.0 camera.
  • In addition to the advanced light microscopy instrumentation the facility boasts other ancillary equipment for sample preparation equipment and cultured cells, . and workstations for data post-processing and analysis.
  • Co-manager Patty Jansma, MS is on-site at this location to support users. Doug Cromey assists in this location one morning a week.

The Life Sciences North location includes:

  • A fully motorized, computer controlled Leica DMI6000 inverted microscope can be used to take brightfield, DIC (20x or above), and polarized light images with a 24bit color camera, or these same imaging modes, as well as four channels of fluorescence, using a 16bit sCMOS greyscale camera.
  • A Zeiss Axio Observer 7 with Apotome III Microscope. This microscope includes an AI sample finder to locate low-contrast samples on a slide or wells in a multiwell plate. It is capable of brightfield color imaging, DIC, POL, and multichannel fluorescence. In widefield mode it can capture up to 5 fluorescence channels, with the Apotome it can capture 4 channels. The Apotome uses a structured illumination technique to provide confocal-like images, including z-stacks, up to about 20um in thickness.
  • A Zeiss Elyra S.1 super resolution fluorescence microscope. This structured illumination microscope (SIM) can be used to image fluorescence markers at twice the resolving power of confocal or deconvolution microscopes. There is a dedicated post-processing workstation with this microscope.
  • An image analysis workstation with commercial (HCImage) and open source (ImageJFIJI, Cellprofiler, Q-Path) image analysis software, as well as proprietary file viewers from all the major optical microscope manufacturers.
  • Co-manager Doug Cromey, MS is on-site at this location to support users.
